| Beta-catenin is one of the key downstream effectors in the Wnt signaling pathway (1). It has been implicated in two major biological processes of vertebrates: early embryonic development (2) and tumorigenesis (3). GSK-3beta may destabilize beta-catenin by phosphorylating it at serines 33, 37 and threonine 41 (4). Serine 45 in beta-catenin is phosphorylated by CK1. This likely serves as a priming site for subsequent phosphorylation by GSK-3 (5-7). Stabilizing mutations of these phosphorylation sites in beta-catenin have been found in many tumor cell lines (8). | | | Catalog # | C2069-51 | | Applications | Suitable for use in Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 1:1000 (Mr 92kD) | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Host | Rabbit | | Source | Human | | Concentration | As reported | | Form | Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol. | | Purity | Purified by Protein A and immunoaffinity chromatography. | | Immunogen | Synthetic phosphopeptide (KLH coupled) corresponding to residues surrounding Ser45 of human beta-catenin. | | Specificity | Detects endogenous levels of betacatenin only when phosphorylated at serine 45. It does not recognize beta-catenin phosphorylated at other sites. Species Crossreactivity: Human, mouse and rat. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
