The alpha-, beta- and gamma-catenins are cytoplasmic proteins mediating the interaction of Ca2+-dependent transmembrane adhesion molecules (cadherins) with the cytoskeletal network. The direct interaction of b-catenin with the cytoplasmic domain of cadherins plays a crucial role for cell-cell adhesion and signal transmission between neighbouring cells. Recent studies indicate that b-catenin may also play a role in tumorigenesis since it forms complexes with the tumor suppressor gene product APC. Beta-catenin directly interacts and constitutively activates transcription factors of the TCF/LEF gene family. Thus it is proposed that b-catenin plays a dual role not only in the maintainance and regulation of cell-cell interactions but also in the regulation of gene activity.

ApplicationsSuitable for use in ELISA, Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested.
Recommended DilutionELISA: 0.05ug/ml
Western Blot: 0.5ug/ml for HRP/ECL detection. Recommended blocking buffer: Casein/Tween-20 based blocking buffer.
Immunocytochemistry: 0.1-1ug/ml
Immunoprecipitation: 1-10ug/ml per 10e6 vanadate treated A431cells.
Optimal dilutions to be determined by the researcher.
Positive Antigen Control (Included)C2069-52C1: Reconstitue in SDS-PAGE sample buffer and incubate at 90ºC for 5 minutes. Recommended for Western Blot at 20ul/lane.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
