ß-Catenin is a 92kD protein that binds to the cytoplasmic tail of E-Cadherin. The cadherins, transmembrane adhesion molecules, are found with catenins at adherens junctions. Deletions in the cytoplasmic domain of ECadherin eliminate catenin binding and result in a loss of cell adhesion. Tyrosine phosphorylation of ß-Catenin can regulate its interaction with critical components of adherens junctions. Both Fer and Fyn kinases phosphorylate tyrosine
142 in vitro. Overexpression of these kinases in epithelial cells disrupts interactions between a- and ß-Catenins. The phosphorylation of tyrosine 142 may act as a switch from the transcriptional to the adhesive role of ß-Catenin. Src family kinases can also phosphorylate tyrosine 654 in the
C-terminal armadillo repeat of ß-Catenin. This phosphorylation regulates ß-Catenin binding to E-cadherin. Thus, site-specific tyrosine phosphorylation of ß-Catenin may regulate specific protein-protein interactions leading to changes in cell adhesion.

Recommended DilutionWestern blotting 1:1000. Membrane was incubated with diluted antibody in 5% non-fat milk, PBS, 0.04% Tween 20 for 1hour at room temperature.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Immunogenß-Catenin synthetic peptide (coupled to carrier protein) corresponding to amino acid residues in the N-terminal region of human ß-Catenin. This human sequence is highly conserved in rat and mouse ß-Catenins.
SpecificityDetects a 92kD protein corresponding to the molecular mass of ß-Catenin on SDS-PAGE immunoblots of Hct116 src transformed cells. Similar results were seen in human endothelial, A431, and HeLa cells.
