Clone Host Grade Applications
Polyclonal Rabbit B
Caspase-3 (CPP32/Yama) is a 32kD member of the group II family of the interleukin-1 beta-converting enzyme (ICE) or cell death effector (CED)-3 cysteine proteases, which is involved in the induction of apoptosis and has been considered to be correlated with apoptosis because of the most downstream enzyme in their apoptosis inducing pathway. Capase-3/CPP32 is immunolocalized in both normal and neoplastic human gastric mucosa (2). The proform of the enzyme has a mitochondrial as well as a cytosolic distribution (3). Within the mitochondrion, pro-caspase-3 is found in a complex with hsp110 and hsp60 (4). Upon activation, procaspase-3 is cleaved into a large subunit p17-p20 depending on the activator, and a smaller p12 subunit (5). Caspase-3 is not expressed in tissue cultured MCF-7 cells (6). T cell apoptosis by cytoskeletal disruption involves a protein synthesis-independent mechanism leading to up-regulation of caspase-3 to its substrate (7). Caspase-3, together with caspase-6, are the major active caspases in apoptotic cells suggesting that activation of caspase-3 is a pivotal event in the execution phase of apoptosis. (2).

Concentration~1mg/ml
FormSupplied as a liquid in PBS, pH7.2, 0.09% sodium azide, 50% glycerol.
PurityPurified by Protein A chromatography.
ImmunogenRecombinant full length human caspase-3 protein with an N-terminal (His) 6 tag.
SpecificityRecognizes human caspase-3. In apoptotic cells, this antibody also detects 20kD and 18kD proteins which correspond to the processed forms of caspase-3. Following the induction of apoptosis and the appearance of these smaller bands, a concurrent reduction in the amount of the procaspase enzyme is noted on immunoblot analysis. A weak ~30kD band may be detected in some samples. This band is not caspase-3 as it is detected in MCF7 cell lysates which lack the caspase-3 protein. Species Crossreactivity: mouse, rat, bovine, canine, chicken, guinea pig, hamster, monkey, porcine, rabbit and yeast.
