Apoptosis, the process of programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms and involves dramatic changes in cellular structure. Disruption of apoptosis has been postulated to contribute to cancer and to autoimmune and degenerative diseases. Apoptosis is induced by a family of death domain (DD) containing cell death receptors and their corresponding ligands. Cell death signals are then transduced by DD, caspase recruitment domain (CARD), and/or death effector domain (DED) containing adapter molecules and by members of the caspase (ICE/CED-3) family of cysteine proteases. Recently identified were FLICE2 and Mch4 (1,2) which are presumed to be at or near the apex of apoptotic signaling pathway (4). They are also known as caspase-10/b and caspase-10/a respectively (4). Caspase-10/b is a long form and caspase-10/a is a shorter form of caspase-10. Caspase-10 has two DEDs that bind to the DED in the adapter molecule FADD and recruits both TNFR1 and Fas (CD95/Apo1) to form complexes with these receptors. Caspase-10 is therefore involved in the Fas and TNFR1 induced apoptosis (1). Caspase-10 cleaves and activates caspase-3,-4,-6,-7,-8, and-9, which are involved in the proteolytic cleavage of many key proteins such as PARP. Cleavage of PARP occurs in many different systems during apoptosis and is the hallmark of programmed cell death (3). Caspase-10 is expressed in many tissues and cell lines (1,2).

the carboxy terminus of human caspase-10/b (FLICE2) (residues 505-521) was synthesized and the peptide coupled to KLH. This amino acid sequence is not found in caspase-10/a (Mch4).
SpecificityDetects a ~59kD protein, corresponding to the apparent molecular mass of caspase-10/b on SDS-PAGE immunoblots, in samples from human, mouse, rat and bovine. An additional band at 40kD is observed in rat and mouse tissue extracts. Does not recognize caspase-10/a (Mch4).
