Procaspase-12 is localized to the endoplasmic reticulum (ER) where it is activated by ER stress and mobilization of intracellular calcium ion store (2). The mechanisms of the activation of caspase-12 by ER stress-induced apoptosis are still unknown. The ER is sensitive to alterations in homeostasis from a variety of different stimuli such as glucose deprivation, peturbation of calcium homeostasis, exposure to free radicals, peturbation of protein folding and the accumulation of malfolded proteins. These events in the ER induce ER stress which elicits two major cellular-protecting responses. One is the attenuation of protein synthesis and the other is the up-regulation of genes encoding chaperones which facilitate the protein folding process in the ER and these responses reduce the accumulation and aggregation of malfolded proteins in the compartments of the cells (3). During the perturbation of protein folding in the ER, the ER stress sensor proteins, IREalpha and IREbeta have been reported to activate C-Jun N-terminal kinases (JNKs) and the activation of JNK requires TNF receptor-associated factor 2 (TRAF2). In unstressed cells, TRAF2 formed a stable complex with procaspase-12. Stimuli induced during ER stress leads to the dissociation of procaspase-12 from TRAF2 and simultaneous oligomerization of procaspase-12 is promoted. The mechanism of caspase-12 activation may be similar to other procaspases such as capase-2,-8, and ñ10 which have been shown to be activated through dimerization/oligomerization mediated through the interaction of their prodomains with adapter molecules (4,5,6,7).
