The a-, b- and g-catenins are cytoplasmic proteins mediating the interaction of Ca-dependent transmembrane adhesion molecules (cadherins) with the cytoskeletal network.
Additionally, b-catenin is a substrate of both receptor and non-receptor tyrosine kinases. Tyrosine 86 and tyrosine 654 are substrates of EGF receptor and src family kinases while tyrosine 142 is a substrate of for tyrosine kinases.

Recommended DilutionWestern Blot: 1ug/ml (HRP/ECL). Recommended blocking buffer: CPPT (0.5% casein, 1% PEG 4000, 1% Polyvinylpyrrolidone (PVP), 0.1% Tween 20, 2X PBS)

Positive cell lysate control includedC2096-64A: Recommended for Western Blot at 20ul/lane.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityRecognizes beta-catenin phosphorylated at Tyr654 and -catenin phosphorylated at Tyr654. Detects bands of ~90kD (beta-catenin) and ~70kD (gamma-catenin). Due to sequence homologies of the phospho-epitope surrounding Tyr654, there may be crossreactions with other proteins (e.g., Annexin A11 at 56kD, desmoplakin at 81kD, TNF-alpha-induced protein 2 at 73kD, ribosomal protein S6 kinase alpha 6 at 90kD). Species Crossreactivity: human, mouse and canine.
