| Caveolin describes a group of 22-24kD integral membrane proteins that are localized to caveolae on the plasma membrane. Caveolae are specialized flask-shaped invaginations of the plasma membrane that are thought to exist in all cell types, but are most prominent in adipocytes, endothelial cells, muscle cells and fibroblasts. Caveolae are thought to play a role in cell signaling and in transcytosis across the plasma membrane. There are three groups of caveolin proteins (caveolin-1, -2 and -3). The gene for caveolin-1 encodes both -1 and -2. Caveolin-3 is expressed primarily in muscle cells. Although caveolin localizes to the plasma membrane, it is insoluble in non-ionic detergents and is believed to be a structural component involved in caveolae assembly. In addition to its structural role, caveolin binds to cholesterol and to G-alpha subunits of heterotrimeric G-proteins. Originally identified with anti-phosphotyrosine antibodies, caveolin is phosphorylated on tyrosine by the Src tyrosine kinase. Phosphorylated caveolin can be detected in v-Src transformed cells, and in cells stimulated with insulin.
