| Catalog # | C2272-73B |
| Applications | Suitable for use in Immunoprecipitation, Immunohistochemistry and Flow Cytometry. Other applications not tested. |
| Recommended Dilution | Immunohistochemistry: Has been reported by an independent laboratory to detect CD21 in 6 micron thick palatine tonsilar sections. |
| Immunoprecipitation: Must use non-denaturing conditions and a different CD21 antibody to detect CD21 in the western immunodetection that follows precipitation. Percent cells with C3d beads specifically bound (%M1) Percent inhibition of C3d-ligand binding No antibody 32.80 0 HB5 (negative control) 31.48 4 Anti-CD21, 3.02 91 Inhibition of C3-Fragment Binding CD21-expressing Raji cells were incubated on ice for 10 minutes with a non-blocking anti-CD21 monoclonal antibody or with Anti-CD21. FITC-labeled agarose microbeads coated with C3d were then added and the reactions continued at room temperature for 20 minutes. Reactions were then terminated and the proportion of cells with one or more beads attached (M1) was measured using a FACScan. |
| FACS Analysis: 2-20ug/ml of this lot detected CD21 in Raji cells suspended in FACS buffer (PBS with 1% BSA and 0.1% sodium azide). Results shown on page two; see also Janz, A., et al. (2000). |
| Inhibition of C3-Fragment Binding: 0.47ug/ml of this lot produced over 90% inhibition of C3d fragment binding to CD21-expressing Raji cells. |
| Optimal dilutions to be determined by the researcher. |
| Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
