| The integrin family of cell adhesion receptors consists of at least 19 membrane-associated heterodimers, composed of an alpha and beta subunit that associate in a non-covalent manner. To date, 16 different alpha-subunits and 8 different beta-subunits have been identified. The structure and functional diversity of the integrin family are based upon the pairing abilities of the individual alpha and beta subunits. Key to these molecular interactions between the integrin receptors and their respective ligands is the recognition of the Arg-Gly-Asp (RGD) sequence, known to be present in the extracellular matrix components fibronectin, vitronectin, collagen, fibrinogen, and von Willebrand factor. The involvement of integrins in vascular proliferation, adhesion, and wound repair has been well documented. Integrin alphaV is a type I membrane glycoptroein with no I (inserted) domain. The alphaV protein is post-translationally cleaved into a heavy and light chain, which is expressed on the cell surface as a disulfide linked polypeptide. | | | Catalog # | C2405-15F | | Applications | Suitable for use in ELISA, RIA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications have not been tested. | | Recommended Dilution | ELISA: 1:500 | | RIA: 1:500 | | Western Blot: 1:250 Recognizes a single band of 150kD when denatured, but not reduced. Under reducing conditions recognizes two major bands of 25 and 27kD, and to a lesser extent the full length 150kD band. This pattern is consitent with the fact that mature alpha V is composed of a disulfide bonded heavy (125kD) and a light (25kD) chains derived from a proteolytically cleaved precursor. | | Immunoprecipitation: 10ul of antibody is sufficient to precipitate alpha V/beta 1 from 5x10e6 cells. Immunohistochemistry: 1:500 Acetone fixed tissue only. | | Optimal dilutions to be determined by the researcher. | | Storage and Stability: | | May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | Not determined | | Form | Supplied as a liquid, 0.05% sodium azide before the addition of glycerol to 40%. | | Purity | Serum | | Immunogen | Synthetic peptide corresponding to the COOH terminal sequence (cytoplasmic domain), aa1022-1034 of human alpha V integrin subunit. | | Specificity | Recognizes human CD51 (Integrin alpha V). The epitope recognized is on the cytoplasmic domain of the protein, and it is thus intracellular. No crossreactivity to alpha 1, alpha 2, alpha 3, alpha 4 or alpha 6 integrin subunits. Species Crossreactivity: goat, equine, porcine, sheep, chicken, hamster, mouse, rat. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
