Endoglin is an endothelial homodimeric membrane antigen containing the tripeptide sequence RGD which is a recognition motif for adhesion receptors of the integrin family. CD105 is a Type I transmembrane protein and exists as an O-and N-glycosylated homodimer. Endoglin is a TGF beta-binding glycoprotein that is most abundantly expressed on endothelial cells in tissues with in ammatory or in ammatory-like characteristics, such as the skin lesions in psoriasis. Proliferating endothelial cells in granulation tissue and in tumors (e.g. melanoma of the skin) also exhibit enhanced expression of endoglin. In addition, a marked staining of syncytiotrophoblasts in placenta is observed. CD105 binds to TGF-b1 and b3 with high affinity but not TGF-b2.

ApplicationsSuitable for use in Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionWestern Blot: 1-2ug/ml for 2hrs at RT.
Immunohistochemistry (Formalin/paraffin): 5-10ug/ml for 60 min at RT. For staining of formalin-fixed tissues, digest sections with Protease XXV at 1mg/ml PBS for 5 minutes at 37°C.
Optimal dilution determined by the researcher.
Positive Control HUVEC cells. Tonsil, lymph node and thymus.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
