| Recommended Dilutions | Western Blot: 1:10. SDS-PAGE under non-denatured conditions. Block blot with 1% skimmed milk for 1h and incubate with C2548-90D2 (0.4ug/ml) in 0.1% BSA/TBS for 1h. Detection of ~130 kD product in spleen extracted in presence of NP-40 or in glucocorticoid-treated human monocytes. (Ref 2,5) |
| Flow Cytometry: 1:10. Permeabilization as well as no permeabilization is used. Block cells with 1% BSA for 30 minutes at 4°C. (Ref 5) |

| Functional Studies: Reduction of CD69 expression by adding 2.5ug/ml C2548-90D2 to 5x10e5 human T-lymfocytes in presence of exogenous CD163. (Ref 2) |
| Immunofluorescence: Transfected CHO-cells were plated on coverslips and fixed in 4% paraformaldehyde for 20 minutes. Cells were stained with 4ug/ml C2548-90D2 for 60 minutes. |
| Immunoprecipitation: 10ug C2548-90D2 was covalently coupled to beads (with 20 mM dimethyl pimelimidate), blocked with 0.2M glycine buffer for 2 hours and incubated with cell lysate for 2 hours (Ref 5). |

| May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
