The anaphase-promoting complex (APC) or cyclosome is a cell cycle-regulated ubiquitin-protein ligase. It is composed of at least 11 subunits that controls exit from mitosis by degrading cyclins. It has been demonstrated that fizzy (also called Fzy or Cdc20) protein is essential for cyclosome activity. In Drosophila, fizzy is required for degradation of A and B type cyclins. Fizzy expression is cell cycle dependent with the peak occurring in M phase. Evidence suggests that fizzy is phosphorylated by Cdk1/cyclin B which inhibits its ability to activate the cyclosome in mitotic HeLa cells. Phosphorylation of fizzy may have a role in keeping the cyclosome inactive during early mitosis and under conditions of mitotic checkpoint arrest.

For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
