| Claudin1, a 22kD integral membrane protein, is a member of Claudin family. It is a critical component of tight junction strands and consists of 2 extracellullar loops, 4 hydrophobic transmembrane domains and a cytoplasmic tail containing PDZ-binding motif. Studies on MDCK cells and knock-out mice have revealed that Claudin-1 plays a vital role in forming tight junctions that serve as a physical barrier preventing free passage of solutes and water through the paracellular spaces. Its role has also been suggested in beta-catenin-Tcf/LEF signaling pathway. Mutations in Claudin-1 have been strongly suggested in NISCH syndrome.
