Claudin-2, a 22kD integral membrane protein, is a member of Claudin family and was discovered along with Claudin-1. It is also a vital component of tight junction strands and consists of 2 extracellullar loops, 4 hydrophobic transmembrane domains and a cytoplasmic tail containing PDZ-binding motif. Studies on MDCK cells suggest that Claudin-2 determines the overall transepithelial membrane resistance properties and increases conductance by forming cation-selective channels in tight junction of epithelial cells. Claudin-2 is differentially expressed in the intestine and has been suggested to be involved in the regulation of intestinal cell monolayer barrier function through immune modulators such as interleukins.
