Claudin-5 is a member of the claudin family. It is a transmembrane protein reported to be a major cell adhesion molecule of tight junctions in brain endothelial cells. Tight junctions are well-developed between adjacent endothelial cells of blood vessels in the central nervous system, and play a central role in establishing the blood-brain barrier (BBB). Claudin-5 is involved in the function of the blood-brain barrier (BBB) and plays a major role in tight junction-specific obliteration of the intercellular space. Mutations in this gene have been found in patients with velocardiofacial syndrome. It is expressed ubiquitously, even in organs lacking epithelial tissues such as the brain, skeletal muscle, and spleen.
