Monoclonal Mouse Affinity Purified FC
CLEC4D (C-type lectin domain family 4 member D), also known as CLCSF8, CLEC-6, and MCL, is a 30kD type II transmembrane (TM) glycoprotein that belongs to the CLR (C-type Lectin Receptor) family of molecules. It is synthesized as a 215aa protein that contains a 17aa N-terminal cytoplasmic domain, a 21aa TM segment, and a 177aa C-terminal extracellular region. The extracellular region shows a short stalk and a 118aa CRD (carbohydrate recognition domain). The nature of its carbohydrate ligand is unknown. CLEC4D is restricted to monocytes/macrophages and serves as an endocytic receptor. Homodimers and homotrimers form on the cell surface. The human CLEC4D extracellular region shares 63% aa sequence identity with the mouse extracellular region.

HybridomaFusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, NS0-derived, recombinant human CLEC4D.
Flow Cytometry:
1. Whole blood monocytes cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. After blocking, add 10ul of C5839-65A to 1-2.5x10e5 cells and incubated for 30 minutes at RT.
3. Remove unbound antibody by washing the cells twice in F4568-10. Note that whole blood requires a RBC lysis step at this point.
4. Resuspend the cells in F4568-10 for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled mouse IgG2B antibody.
Storage and StabilityMay be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Freezing APC conjugates will result in a substantial loss of activity. APC conjugates are sensitive to light.
