| Collagens are highly conserved throughout evolution. They are characterized by an uninterrupted "Glycine-X-Y" triplet repeat that is a necessary part of the triple helical structure. For these reasons it is often extremely difficult to generate antibodies with specificities to collagens. The development of type specific antibodies is dependent on non-denatured three-dimensional epitopes. |

| • A0030: ABTS Stable Liquid Substrate (2,2’ -Azino-bis- |
| [3-ethylbenzthiazoline-6-sulfonic acid]) |
| Western Blot (Non-denaturing, non-dissociating) | 1:2500-1:5000 |
| Immunohistochemistry (frozen tissue sections) | 1:25-1:100 |
| Some class speci c anti-collagens may be speci c for three-dimensional epitopes which may result in diminished reactivity with denatured collagen or formalin- xed, paraf n embedded tissues. |
| Optimal dilutions to be determined by the researcher. |
| Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
| CAS Number | n/a |
| Clone Type | Polyclonal |
| Host | Rabbit |
| Source | Human |
| Concentration | ~0.5mg/ml |
| Form | Supplied as a liquid in 0.125M sodium borate, 0.075M sodium chloride, 0.005M EDTA, pH 8.0, 0.01% sodium azide before the addition of glycerol to 40%. No stabilizing proteins added. |
| Purity | Purified by immunoaffinity chromatography. |
|
| Immunogen | Collagen Type II from adult human knee cartilage and bovine nasal cartilage. Collagen Type II was extensively purified by limited pepsin digestion and selective salt precipitation. This preparation results in a native conformation of the protein. |
| Specificity | Recognizes Type II collagens. Species crossreactivity: most mammals. Crossreactivity with Type I, III, IV, V or VI collagens is negligible. Non-speci c crossreaction of anti-collagen antibodies with other human serum proteins or non-collagen extracellular matrix proteins is negligible. |
