ApplicationsSuitable for use in ELISA, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionELISA: 1:4000-1:8000. Assayed against 1ug of Collagen IV in a standard sandwich ELISA using S7973-97A: Streptavidin (HRP) and A0030: ABTS Stable Liquid Substrate 1.46mM/Liter for HRPO for 30 minutes at RT.
Immunohistochemistry (Frozen tissue): 1:50-1:200
Western Blot: 1:5000-1:10,000
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityRecognizes Collagen Type IV. Typically less than 1% crossreactivity against other types of collagens was detected by ELISA against puri ed standards. Some class speci c anti-collagens may be speci c for three-dimensional epitopes which may result in diminished reactivity with denatured collagen or formalin- xed, paraf n embedded tissues. Reacts with most mammalian Type IV collagens and has negligible crossreactivity with Type I, II, III, V or VI collagens. Non-speci c crossreactivity with other human serum proteins or non-collagen extracellular matrix proteins is negligible.
