| C1r is a plasma protein and forms part of the C1 pentamer, the first component of the classical complement pathway. The C1r dimer binds to two C1s molecules in a Ca2+-dependent manner. This tetramer then binds to C1q to form the C1 pentamer. C1r is a serine protease and on the binding of C1 to an immune complex, C1r self activates. Activated C1r (CTr) in turn activates C1s, the other serine protease component of C1. Activated C1 in turn activates C4 and C4bC2 and is inhibited by C1-inactivator.Normal plasma concentrations of C1r are around 34 mg/L. Unactivated C1r consists of a single polypeptide chain of MW 173kDa.
