C5 is involved in the activation of the lytic pathway within the complement system which is an important factor in innate immunity. The complement pathways can be divided into the activation pathways and lytic pathway. The activation pathways lead, via C3, to the cleavage of the fifth complement component C5. C5a induces smooth muscle contraction, increases vascular permeability, causes degranulation of mast cells and basophils, and release of lysosomal enzymes. In addition C5a stimulates the directed migration of neutrophils, eosinophils, basophils and monocytes. Recent studies indicate the modulation of the acute-phase response in liver and an overall immune response by inducing synthesis of cytokines such as TNF-alpha IL-1beta, IL-6 and IL-8.

Applications
Suitable for use in ELISA, Western Blot, Immunohistochemistry and Functional Studies. Other applications not tested.
Recommended Dilutions
Western Blot: 1:50 Immunohistochemistry (Paraffin): 1ug/ml. No pretreatment was used on human tissue sections. Human granulocytes were used as a positive control. Immunohistochemistry (Frozen): Tissues sections were fixed in Histochoice MB and blocked with DakoCytomation phosphatases. Pretreatment with levamisole solution was used to quench endogenous phosphatases. Drusen eye sections were used as a positive control and sections lacking the antibody were used as the negative. Functional Studies: Functions as a neutralizing antibody blocking the interaction between C5a and its receptor. Functionally tested by competitive binding studies using C5a and Bt2-cAMP differentiated U937 cells. The biological activity can be defined as the ability of C7850-24H to displace C5a from U937 cell surface. Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
