| Two distinct members of the Cullin gene family- Cul3 and Cul11- have been implicated in the ubiquitin-mediated destruction of mammalian cyclin E. Cul3 may be a core component of the E3 ubiquitin ligase complex that targets free cyclin E for destruction. Cul3 physically associates with cyclin E in mammalian cells, as would be expected if Cul3 was a component of the cyclin E ubiquitin ligase. Overexpression of Cul3 stimulates the accumulation of cyclin E-ubiquitin conjugates in a phosphorylation-independent manner, thereby demonstrating a critical link between Cul3 and the cellular cyclin E destruction machinery. Cul3 is likely to be required for destruction of cyclin E. However, it seems unlikely that the development defects are solely due to cyclin E accumulation, as Cul3 complexes will likely regulate the levels of other proteins in addition to cyclin E. Human Cul3 is a member of the Cullin family of E3 ubiquitin-protein ligases. Cul3 was found to bind to cyclin E but not to cyclin E-cdk2 complexes in mammalian cells, and that overexpression of Cul3 increased ubiquitination of cyclin E but not other cyclins. The SCF pathway, which targets cyclin for ubiquitination on the basis of their phosphorylation state, and the Cul3 pathway, which selects cyclin E for ubiquitination on the basis of its assembly into cdk complexes, may be a complementary way to control cyclin abundance. | | | Catalog # | C7949-21 | | Applications | Suitable for use in Western Blot and Immunoprecipitation.
