CXCR3 is a cell surface chemokine receptor that belongs to the family of G protein linked, seven transmembrane domain containing receptors. With the cloning of this molecule, it was identified as the specific receptor for two CXC chemokines: gamma interferon induced protein and monokine induced by interferon gamma. The CXCR3 receptors appear to have affinity for CXC chemokines that do not contain the conserved ELR motif at their amino terminus and as a result IL-8 and platelet factor 4 do not bind to this receptor. It shares approx. 40% monology with the CXCR1 and CXCR2 receptors and approx 35% homology with the first five CCR receptors. In spite of the fact that the two ligands for CXCR3 are members of the a-chemokine family, which has historically linked with chemotactic activity of neutrophils, IP10 and MIG, are the first molecules to target lymphocytes. This is consistent with the mRNA expression pattern of CXCR3 molecules, where it was found on activated T, B, and NK cells but not on monocytes, resting T cells, or neotrophils. Of clinical interst is the demonstration of CXC3 expression on cells from B cell chronic lymphocytic leukimeias and splenic marginal zone lymphomas.
