| CXCR3 is a G protein-coupled receptor with selectivity for three chemokines, termed IP10, Mig and I-TAC. The protein is mainly expressed on activated Th1 cells, B cells and natural killer cells. It is activated by the interferon-gamma-inducible chemokines CXCL9, CXCL10 and CXCL11, with CXCL11 having the highest affinity. Upon activation, CXCR3 activates pertussis toxin-sensitive G proteins of the Galphai class and mediates, among others, chemotaxis, calcium flux and activation of kinases such as p44/p42 MAPK and Akt. The protein and its ligands are upregulated in a wide variety of inflammatory diseases, implying a role for CXCR3 in serious disorders (e.g. rheumatoid arthritis, multiple sclerosis, transplant rejection, atherosclerosis and inflammatory skin diseases) and thus has attracted considerable attention as a drug target.
