CXCR6, is a G protein-coupled chemokine receptor expressed on activated T cells and tumor-infiltrating lymphocytes. It binds soluble CXCL16 and membrane-anchored CXCL16 expressed on dendritic cells and macrophages.

Recommended DilutionFlow Cytometry: 10ul labels 1x10e6 cells

HybridomaMyeloma cells with B cells from Balb/c mice.
Flow Cytometry Protocol1. Cells may be Fc-blocked with 1ug of mouse IgG/10e5 cells for 15 minutes at room temperature. Do not wash excess blocking IgG from this reaction.
2. After blocking, 10ul of conjugated antibody was added to 1x10e6 cells and incubated for 30 minutes at room temperature.
3. Unbound antibody was removed by washing the cells twice in F4568-10 Flow Cytometry Staining Buffer (1X). Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Mouse Lyse Buffer.
4. The cells were resuspended in Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with CFS-labeled rat IgG2B antibody. This procedure may need to be modified, depending upon the cell type and final utilization. Individual users may need to titrate to determine the optimal reagent amount for their specific use.
Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Carboxyfluorescein conjugates are sensitive to light.
