| Cyclins are proteins that regulate cell cycle progression by interacting with cdc2 kinase and/or related kinases. Human cyclins have been grouped into 5 types, A through E, based largely upon sequence similarity. Besides serving as cell cycle regulators, D-type cyclins have been reported to be able to interact with several transcription factors and modulate their transcriptional activations. Cyclin D1, a member of the D-type cyclins, is an important regulator of G1 to S-phase transition in numerous cell types from diverse tissues. Binding of cyclin D1 to its kinase partners, the cyclin dependent kinases 4 and 6 (CDK46) results in the formation of active complexes that phosphorylate the Retinoblastoma tumor suppressor protein (RB). It is an oncogene amplified or over-expressed in more than 50% of all breast cancers. Recent work reveals the role of Cyclin D1 in regulation of cellular metabolism, fat cell differentiation and cellular migration.
