Cyclins were originally identified as proteins whose levels fluctuated during the cell cycle. Subsequent studies have shown that the cyclins control entry into different phases of the cell cycle by regulating the activity of cyclin-dependent kinases (CDKs). Mitotic or G2 cyclins regulate entry into M phase, whereas G1 cyclins regulate entry into S phase. Cyclin D1, a G1 cyclin, is essential for control at the G1/S transition (START in yeast), as it regulates the activity of CDK4 and CDK6 protein kinases. Translocation of the gene encoding Cyclin D1, the bcl-1 oncogene, is associated with human B-lymphocytic malignancy, suggesting that alteration of cell cycle progression may be oncogenic.

Application	Immunoblot Analysis 2: At 0.5–2ug/ml, this antibody detected Cyclin D in a RIPA lysates of human A431 cells, mouse 3T3 fibroblasts.
Positive Antigen Control	EGF-stimulated A431 cell lysate. Use 20ug per lane.
Immunoprecipitation	5ug of antibody immunoprecipitated Cyclin D from a RIPA lysate of human A431 cells. An H1 kinase assay can be performed with the immunoprecipitate, as reported previously.
Immunocytochemistry	At 10ug/ml, a previous lot of antibody detected Cyclin D in cells fixed with 4% paraformaldehyde/0.1% Triton X-100. Immunoblot Analysis EGF-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Cyclin D (2ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.

Immunogen	Synthetic peptide corresponding to the 11 C-terminal amino acids (residues 285–295; sequence: CTPTDVRDVDI) of human Cyclin D1.
Specificity	Recognizes p36 Cyclin D1 and crossreacts with p34 Cyclin D2, which has a highly homologous epitope. No crossreactivity with Cyclin A or Cyclin B. Shows reactivity with a 28kDa band that can competed away by pre-incubating antibody with the peptide immunogen. Also detect a nonspecific band at 21kDa. Species crossreactivity: mouse and rat.
