Cyclins are proteins that regulate cell cycle progression by interacting with cdc2 kinase and/or related kinases. Human cyclins have been grouped into 5 types, A through E, based largely upon sequence similarity. Progression through the G1 phase of the cell cycle and entry into S phase in mammalian cells are controlled by a number of proteins, including the cdc2-related kinases cdk2, cdk4 and cdk6, their regulatory subunits, the D and E type cyclins. Cyclin E forms complexes specifically with cdk2 and activates its serine-threonine kinase activity shortly prior to entry into S phase, thus regulates the G1/S transition. It was found to associate with, and be involved in, the phosphorylation of NPAT protein (nuclear protein mapped to the ATM locus), which participates in cell-cycle regulated histone gene expression and plays a critical role in promoting cell-cycle progression in the absence of pRB. The existence of a 43kD splice variant of human cyclin E, termed cyclin ES, which lacks 49aa within the cyclin box compared with the known full-length 48kD cyclin E has been reported. It is highly expressed in testis and placenta and low levels in bronchial epithelial cells. Overexpression of this gene has been observed in many tumors, which results in chromosome instability, and thus may contribute to tumorigenesis.
