| Cyclin E is a regulatory subunit of cdk-2 and is thought to be rate limiting for the G1/S transition during the mammalian cell cycle. Multiple isoforms of cyclin E are expressed only in tumors but not in the normal tissues, suggesting a post-transcriptional regulation of cyclin E. In vitro analyses indicated that these truncated variant isoforms of cyclin E are able to phosphorylate histone H1. Qualitative and quantitative alterations in cyclin E protein have been implicated as indicators of worse prognosis in various cancers. For biochemical applications including co-precipitation of cdk2. |

| Applications | Suitable for use in Immunofluorescence, Immunoprecipitation, Immunohistochemistry, Kinase Assays, and Western Blot. Other applications not tested. |
| Recommended Dilution | Immunoprecipitation: (Co-precipitates cdk2, Use Protein A; Ab at 10ug/mg protein lysate) |
| Western Blot: Ab at 5-10ug/ml for 2hrs at RT. |
| Immunohistochemistry (formalin/paraffin): 10-20ug/ml for 30 minutes at room temperature. |
| Note: Staining of formalin/paraffin tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. |
| Optimal dilutions to be determined by the researcher. |
| Cellular Localization | Nuclear |
| Positive Control | MAD109 and SKBR3 cells. Breast carcinoma or placenta. |
| Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C or colder. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
