| Cystatins are cysteine protease inhibitors constituting a superfamily of evolutionary related proteins, composed of at least one 100-120aa residue domain with conserved sequence motifs. The single-domain human members of this superfamily are of two major types, type 1 and type 2. Cystatin A belongs to family 1 of the cystatin superfamily. It is an inhibitor of lysosomal cysteine proteinases cathepsin B, H, L and S. It was originally isolated from polymorphonuclear granulocytes, but it has also been isolated from the spleen, liver, and epidermis. The gene encoding the protein consists of three exons and two introns. The protein consists of 98aa residues with a molecular mass of 11006D, lack disulfide bridges, and is synthesized without signal peptides. Recent reports reveal that cystatin A is identical to keratolinin, one of the precursor proteins of cornified cell envelope (CE), which is formed during terminal differentiation of keratinocytes.
