Cytochrome C/Apaf-2 (apoptosis protease activating factor-2) is well known for its role in electron transport. During apoptosis (programmed cell death), Cytochrome C is released from the mitochondria into the cytosol where it binds to Apaf-1. Apaf-1 then activates caspase 9. Caspase 9 activation triggers other downstream caspase events in the proteolytic cascade. The mechanism by which Cytochrome C is released from the mitochondria and its function in Caspase 9 activation is still unclear.

ApplicationsSuitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested.
Recommended DilutionWestern Blot Analysis: 1:500-1:2000 detects Cytochrome C in RIPA lysates from A431 cells. EGF-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Cytochrome c (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Immunohistochemistry: Reported to stain rat spine cord sections.
Optimal dilutions to be determined by the researcher.
Positive Antigen ControlEGF-stimulated A431 lysate. Use 20ug per lane for minigels.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
