| Catalog # | C9095-17F | | Source | Insect cell microsomes prepared from cells that were infected with a baculovirus containing the cDNA for human CYP2C9 and rabbit cytochrome P450 oxidoreductase. The CYP2C9 allele expressed is the most common in human populations studied thus far. The allele contains Arg, Ile, and Gly at positions 144, 359, and 475 respectively. | | Specific Content of Cytochrome P450 | 172pmol spectral P450/mg Protein. | | Cytochrome P450 reductase activity | 1.2088umol of Cytochrome c reduced per minute per milligram of protein. | | Diclofenac hydroxylase activity | 5.9 ± 0.881nmol/min/mg of protein. | | Diclofenac hydroxylase reactions were performed for 10 minutes at 37°C. Reaction were performed in the presence of 0.1M Tris HCl, pH 7.5, 100uM diclofenac, 2mM NADPH, and at a final protein concentration of 250ug/ml. The reactions were stopped by addition of 20ul acetonitrile/glacial acetic acid (96 | 4). The reactions were centrifuged for 10 min. at maximum speed in a microfuge. 10ul of the supernatant was injected onto a 4.5mm x 70mm x 3 micron C18 HPLC column. | | Storage and Stability | Store at -80°C.
