SourceInsect cell microsomes prepared from cells that were infected with a baculovirus containing the cDNA for human CYP2C9 and rabbit cytochrome P450 oxidoreductase. The CYP2C9 allele expressed is the most common in human populations studied thus far. The allele contains Arg, Ile, and Gly at positions 144, 359, and 475 respectively.

Diclofenac hydroxylase activity5.9 ± 0.881nmol/min/mg of protein.
Diclofenac hydroxylase reactions were performed for 10 minutes at 37°C. Reaction were performed in the presence of 0.1M Tris HCl, pH 7.5, 100uM diclofenac, 2mM NADPH, and at a final protein concentration of 250ug/ml. The reactions were stopped by addition of 20ul acetonitrile/glacial acetic acid (964). The reactions were centrifuged for 10 min. at maximum speed in a microfuge. 10ul of the supernatant was injected onto a 4.5mm x 70mm x 3 micron C18 HPLC column.
Storage and StabilityStore at -80°C. Avoid repeated freeze/thaw cycles as this will decrease the activity of the enzyme.
