| This gene, CYP3A4, encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism andsynthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmicreticulum and its expression is induced by glucocorticoids and some pharmacological agents. This enzyme is involved in the metabolism of approximately half the drugs which are are used today, including acetaminophen, codeine, cyclosporin A, diazepam and erythromycin. The enzyme also metabolizes some steroids and carcinogens. This gene is part of a cluster of cytochrome P450 genes on chromosome 7q21.1. Previously another CYP3A gene, CYP3A3, was thought to exist; however, it is now thought that this sequence represents a transcript variant of CYP3A4. | | | Catalog # | C9095-18F | | Applications | Suitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested. | | Recommended Dilution | Western Blot: 1:1000-1:2000. Using a microsomal membrane fractions, detects a band at ~54kD although other bands may be present as well. Successfully detects CYP3A4 on LICOR fluorescent westerns. | | Immunohistochemistry: 1:200-1:400 on 10% formalin fixed, paraffin embedded tissues. Microwave-citric acid antigen recovery is suggested. | | Optimal dilutions to be determined by the researcher. | | Control | Human liver microsomes | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | Not determined | | Form | Supplied as a liquid.
