| NADPH-cytochrome P450 reductase is a microsomal component that is essential for transferring reducing equivalents from NADPH to the microsomal heme oxygenase isozymes, HO-1 and HO-2, to the various cytochrome P450 isozymes and to cytochrome c and b5. The reductase is inducible in the liver, phenobarbital and 3-naphthoflavone are good inducers of the enzyme. The rat enzyme has an apparent molecular weight of 78kD and contains both FAD and FMN. Nitric oxide synthase is the other known enzyme that also contains FAD and FMN. The reductase has been isolated from different species and a close range of apparent molecular weights of 76 to 81kD. The amino acid sequence of the reductase from several species has been deduced from cDNA. The enzyme has a short hydrophobic N-terminus that serves to anchor the reductase to the endoplasmic reticulum and nuclear envelope; the reductase without the hydrophobic portion retains its ability to transfer electrons to cytochrome c, but not to cytochrome P450. The 70kD hydrophilic region contains the flavin and NADPH binding domains. The purified enzyme in the presence of a detergent, such as Triton X-100 (0.02%), can reconstitute heme degradation activity of purified or expressed heme oxygenase isozymes(1,3–8). | | | Catalog # | C9095-24 | | Applications | Suitable for use in Western Blot.
