Among the cytoplasmic intermediate filaments (IF) proteins, keratins make up the largest family and are expressed specifically in epithelial cells in a cell-specific manner. Keratins include more than 20 unique gene products (termed K1-K20) that are divided into type I (K9-K20) and type II (K1-K8). Most epithelial cells express at least one type I and one type II keratin as their predominant IF protein complement in an epithelial cell-specific manner. The protein encoded by this gene is a type I cytokeratin. It is expressed in abundance in nail bed, hair follicle, sebaceous glands, and other epidermal appendages. Mutations in this gene lead to Jackson-Lawler type pachyonychia congenita and steatocystoma multiplex. It maps to 17q12-q21 region of human chromosome. It may be a marker of basal cell differentiation in complex epithelia and therefore indicative of a certain type of epithelial stem cells.
