The cytokeratins are a family of intermediate filaments fond in almost all epithelial cells. They are divided into four subclasses depending on their patterns of reactivity and electrophoretic characteristics and are also arranged I a numbered series of 20 or more acoring to molecular weight and isoelectric point (Quinlan et al. 1985). Types 5 and 6 of cytokeratin (CK5/6) are in high frequency in mesothelial cells, as opposed to cells of adenocarcinoma. CK5/6 has been shown to be a reliable marker for mesothelioma and squamous cell carcinoma of the lung and Bowen's disease. It does not react with pulmonary adenocarcinoma.

Recommended DilutionImmunohistochemistry (frozen or paraffin sections): 1:50-1:100
Western Blot: Recognizes a 58kD and 56kD band corresponding to cytokeratin 5 and 6. Cytoskeletal preparations of epidermis.
Optimal dilution determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

ImmunogenPurified cytokeratin 5
SpecificityRecognizes human cytokeratin 5. Demonstrates crossreactivity with cytokeratin 6. Demonstrates weak crossreactivity with cytokeratin 4 on immunoblots. Does not crossreact with cytokeratins 1, 7, 8, 10, 13, 14, 18 or 19. Recognizes basal cells and a part of stratum spinosum in the normal pavement epithelium.
