DACT1 is a 836aa protein belonging to the Dapper family. It antagonizes Wnt signaling by promoting DVL2 (Dishevelled) degradation via a lysosome inhibitor-sensitive and proteasome inhibitor-insensitive mechanism and the interaction is formed between the DEP (Dishevelled, Egl-10, and pleckstrin) domain of Dvl and the central and C-terminal regions of DCT1. Wnt signaling plays a key role in embryogenesis and its dysregulation leads to cancer development. It binds to DVL2 and causes the degradation of CTNNB1/beta-catenin, thereby enhancing the transcriptional activation of target genes of the Wnt signaling pathway. DCT1 is required for normal notochord formation and is downregulated in hepatocellular carcinomas (HCC). It is expressed in liver and various other tissues.
