| DARPP-32 (dopamine and cyclic AMP regulated phosphoprotein, relative MR 32kD) is a cytosolic protein highly enriched in mediumsized spiny neurons of the neostriatum (1). Dopamine stimulates phosphorylation of DARPP-32 through D1 receptors and activation of PKA. PKA phosphorylation of DARPP-32 at threonine 34 converts it into an inhibitor of protein phosphatase 1 (1). DARPP-32 is converted into an inhibitor of PKA when phosphorylated at threonine 75 by cyclin-dependent kinase 5 (cdk5) (2). Mice containing a targeted deletion of the DARPP-32 gene exhibit an altered biochemical, electrophysiological and behavioral phenotype (3). DARPP-32 is a bifunctional signal transduction molecule that controls serine/threonine kinase and serine/threonine phosphatase activity depending on the phosphorylation state of the protein (2). | | | Catalog # | D1075 | | Application | Suitable for use in Phosphatase assays. | | Other applications not tested. | | Recommended Dilution | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | Source | Recombinant human DARPP-32, expressed in bacteria with a His-Tag linker. | | Purity | ~90% by SDS-PAGE. The Recombinant DARPP-32 His fusion protein preparation shows two bands of 38kD and 26kD, the latter representing a form lacking ~10–12kD from the carboxy terminus. Both bands are excellent substrates for phosphorylation. | | Concentration | ~0.5mg/ml | | Form | Supplied as a liquid in Tris-buffered saline, 40% glycerol. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
