| DARPP-32, a dopamine (DA) and cAMP-regulated ~32kD phosphoprotein that is associated with dopaminoceptive neurons bearing D-1 receptors in the basal ganglia,1 inhibits protein phosphatase I when it is phosphory on Thr34. In contrast, when DARPP-32 is phosphorylated on Thr75, the protein acts as an inhibitor of PKA. Phosphorylation of DARPP-32 is thought to play a critical role in the regulation of dopaminergic neurotransmission2. In addition, the activity of DARPP-32 is also thought to play important roles in the actions of alcohol, caffeine and Prozac®. | | | Catalog # | D1075-03A | | Applications | Suitable for use in Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 1-3ug/ml | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at -20°C for short-term only. For long-term storage, store at -70°C. Aliquots are stable for at least 12 months at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Host | Rabbit | | Concentration | 10 mini-blots/100ul | | Form | Supplied as a liquid in 10mM Hepes, pH 7.5, 150mM NaCl, 0.1mg/ml BSA, 50% glycerol. | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | Synthetic peptide derived from amino acids surrounding the phosphorylated Thr34 residue of DARPP-32. | | Specificity | Reacts with human, mouse, and rat DARPP-32 proteins phosphorylated at Thr34. On Western blots, it identifies the target band at ~32 kD. Reactivity has been confirmed with rat caudate slices incubated in the presence of forskolin. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
