DLL1 is the human homolog of the Drosophila Notch ligand, Delta. It is a type I transmembrane protein with a DSL (Delta, Serrate, Jagged) domain followed by 8 EGF-like domains in its extracellular region and a short cytoplasmic region. Notch-DSL signaling plays an important role in the differentiation processes of many tissues.

ApplicationsSuitable for use in Flow Cytometry. Other applications not tested.
Recommended DilutionFlow Cytometry: 10ul labels 1x10e6.
Optimal dilutions to be determined by the researcher.
HybridomaFusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, NSO-derived recombinant human Delta Like Protein 1.
Flow Cytometry:
1. T98G cells. may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. After blocking, add 10ul of D2727-70C to 1x10e6 cells and incubated for 30 minutes at RT.
3. Wash the cells twice in F4568-10 Flow Cytometry Staining Buffer. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer.
4. Resuspended the cells in F4568-10 Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled mouse IgG2B antibody. This procedure may need to be modified, depending upon the cell type and final utilization. Individual users may need to titrate to determine the optimal reagent amount for their specific use.
Storage and StabilityMay be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C. Stable for 12 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Freezing APC conjugates will result in a substantial loss of activity. APC conjugates are sensitive to light.
