Desmin is a 53 kD cytoplasmic protein, consisting of a nonhelical N-terminal headpiece, a C-terminal tailpiece, and an alpha-helical middle domain of approximately 300 amino acids. Desmin shares 30-70% amino acid homology with other intermediate filaments, but the antibody does not stain other filament proteins such as vimentin, keratin, glial fibrillary acidic protein, and neurofilament protein. Desmin is a marker for myogenic differentiation; it stains smooth muscle cells, myocardium, and striated muscle cells. It does not stain epithelial and myoepithelial cells, endometrial stroma, ovarian stroma, lymphoid tissue, osteocytes, chondrocytes, or mesothelial cells. In neoplastic tissue, desmin stains leiomyoma (12/13 cases), rhabdomyoma (6/6 cases), rhabdomyosarcoma (31/31 cases), and leiomyosarcoma (18/26 cases). Alveolar and embryonal rhabdomyosarcomas consistently stain positive with desmin. In addition to myogenous cells, desmin also stains myofibroblasts. Desmin focal staining may be seen in myofibroblastic differentiated tumors, including fibromatosis and dermatofibrosarcoma protuberans. Myofibroblasts with positive desmin staining may be observed in malignant fibrous histiocytoma, inflammatory pseudotumors, and postoperative spindle cell nodules.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile (40-50%) glycerol, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
