DNA polymerase b comprises an amino-terminal 8-kD domain and a carboxyterminal 31-kD domain. The N-terminal ssDNA binding domain has a deoxyribose phosphodiesterase activity while the C-terminal domain has a nucleotidyltransferase activity. Mammalian DNA polymerase b, a DNA repair polymerase, is constitutively expressed in cultured cells, but treatment of cells with the DNA-alkylating agents such as N-methyl-Ní-nitro-N-nitrosoguanidine (MNNG) or methyl methanesulfonate up-regulates beta-pol level. DNA polymerase b fills single nucleotide gaps in DNA produced by the base excision repair pathway of mammalian cells.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at-20°C or colder. Aliquots are stable for at least 12 months at-20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
