Specifications

Dipeptidyl peptidase-4 (DPP4), also known as adenosine deaminase complexing protein 2, is a serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides. DPP4 plays a key role in glucose metabolism, immune regulation, signal transduction and apoptosis. The DPP4 Fluorescent Activity Assay kit is designed to measure DPP4 activity using purified DPP4 or cell extracts containing DPP4. It comes in a convenient 96-well format, with purified DPP4 enzyme, DPP4 substrate, and DPP4 assay buffer for 100 enzyme reactions. The key to the DPP4 Fluorescent Activity Assay is the specific, fluorogenic substrate. Using this kit, only one simple step on a microtiter plate is required for DPP4 reactions. The DPP4 fluorometric substrate is incubated with a sample containing DPP4 enzyme to produce a fluorophore that can then be measured using a fluorescence reader.

Kit Components1. DPP4 human recombinant enzyme: 1x1ug. Store at -70°C.
2. DPP4 assay buffer: 1x10ml. Store at -20°C.
3. Fluorogenic DPP substrate 1 in DMSO (0.5mM): 1x100ul. Store at -70°C.
4. AMC Fluorescent standard (50uM): 1x500ul. Store at -20°C.
5. Black, low binding NUNC black microtiter plate: 1 plate. Store at room temperature.
APPLICATIONSSuitable for studying enzyme kinetics and screening small molecular inhibitors for drug discovery and HTS applications. Other applications not tested.
Storage and StabilityStore buffer and standard at -20°C. Store enzyme and substrate at -70°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
