DPL2 (DPP10), a novel 796-amino-acid protein, with significant sequence identity to DPP4 (32%) and DPP6 (51%) respectively. is a member of the dipeptidyl peptidase IV (DPIV) gene family. DPL2 encodes a single-pass type II membrane protein that is a member of the S9B family in clan SC of the serine proteases. The DPP10 gene is located on the long arm of chromosome 2 (2q12.3-2q14.2). Alternate transcriptional splice variants, encoding different isoforms, DPL2-s, DPL2-l have been characterized. DPL2 mRNA is expressed mainly in the Human brain and pancreas. DPL2 lacks active-site serine residue being replaced by a glycine residue required to confer dipeptidyl peptidase activity and has instead evolved features that enable it to act as an important component of voltage-gated potassium channels. Mutations in DPL2 gene have been associated with asthma.
