DM-GRASP (BEN, SC1) is an adhesion molecule that was originally described as a marker in the developing central and peripheral chicken nervous systems, where it was expressed by subsets of motor and sensory nervous during phases of axonogenesis, neurite elongation, axonal fasciculation, and synapse formation (2). DM-GRASP belongs to the CAM immunoglobulin superfamily of adhesion molecules, and exists as a cell surface glycoprotein of ~90-105kD (1). Structurally, DM-GRASP is related to MUC18, a human melanoma marker, but sequence identity between the two proteins is only ~25% (1). Homophilic GRASP-GRASP interactions between a growing neuron and a substrate appear to be an essential component of DM-GRASP’s neurite extension-stimulating activity (4). In chickens, levels of DM-GRASP expression on neurons are highest during the period of active neuronal extension, and disappear after they have reached their targets (5). A similar pattern has been observed during chick neuromuscular development; both motoneurons and their target, the myoblasts, express DM-GRASP during early embryonic development, then demonstrate restricted protein expression at neuromuscular
contacts as soon as postsynaptic acetylcholine receptor clusters are observed (6).
