| p62Dok was first identified as a 62 kD RasGAP-binding protein in v-Abl transformed B cells and in Bcr-Abl expressing cells. It is a novel protein with features of a signaling molecule. Association of p62Dok with GAP correlates with its tyrosine phosphorylation. It is rapidly tyrosine-phosphorylated upon activation of the c-Kit receptor, implicating it as a component of a signal transduction pathway downstream of receptor tyrosine kinases. The protein contains a putative pleckstrin homology domain at the amino terminus and ten PXXP SH3 recognition motifs. Thefunctional role of the protein is implicated in the regulation of a wide range of physiological responses, including negative regulation of cytokines, enhanced cell migration, filopodia extension and tumor suppression.
