| p56Dok-2 was initially identified as one of several tyrosine phosphorylated proteins derived from bone marrow taken from patients with chronic myeloid leukemia. The protein consists of a predicted N-terminal pleckstrin homology domain as well as several other features of a signaling molecule, including 13 potential tyrosine phosphorylation sites, six PXXP motifs, and the ability to bind to p120RasGAP. It is shown to be 35% identical to p62Dok-1, a recently identified RasGAP binding protein from CML cells. p56Dok-2 is implicated in the regulation of a wide range of physiological responses, including negative regulation of cytokines, enhanced cell migration, filopodia extension and tumor suppression.
