| EDG1 is a member of the endothelial differentiation gene family. It is a high affinity G protein-coupled receptor for the bioactive lipid, Sphingosine-1-phosphate. EDG-1 signaling regulates endothelial cell survival, cytoskeletal remodeling, chemotaxis and angiogenesis. | | | Catalog # | E2208-01D | | Applications | Suitable for use in Flow Cytometry. Other applications not tested. | | Recommended Dilutions | Flow Cytometry: 10ul labels 10e6 human EDG-1/S1P1-transfected CHO-S cells. | | Optimal dilutions to be determined by the researcher. | | Flow Cytometry Protocol | 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction. | | 2. After blocking, 10ul of E2208-01D was added to up to 1x10e6 cells and incubated for 30 minutes at RT. | | 3. Unbound antibody was removed by washing the cells twice in Flow Cytometry Staining Buffer (Catalog # F4568-10). Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer. | | 4. The cells were resuspended in F4568-10 for final flow cytometric analysis. As a control for analysis, cells in a separate tube should be treated with APC-labeled mouse IgG2b antibody. | | Storage and Stability | May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
