| Edg-1 is a heterotrimeric orphan receptor for lysosphingolipid sphingosine 1-phosphate (S1P) and belongs to the G-protein coupled receptor 1 family with seven putative transmembrane domains. Human Edg-1 is a highly inducible and abundant endothelial cell GPR that helps in the regulation of differentiation of endothelial cells. S1P-induced endothelial cell migration requires the PKB:AKT1 mediated phosphorylation of Edg-1. Activation of Edg-1 induces cell-cell adhesion and it also helps in modulating intracellular Ca2+ homeostasis. It is rapidly induced when endothelial cells are treated with the tumor promoter phorbol 12-myristate 13-acetate (PMA) and super induced in the presence of cycloheximide. Over expression of Edg-1 leads to cell-cell aggregation, enhanced expression of cadherins and formation of well-developed adherens junctions.
