Endothelial Cell Differentiation Gene-7, or EDG-7, encodes a 40kD protein belonging to a family of G-Protein Coupled Receptors (GPCR’s), which bind lysophospholipids. To date, 8 members of the EDG receptor family have been identified. These receptors are suggested to have a role in modulation of growth-related effects including cell proliferation and differentiation as well as protective, anti-apoptotic effects. EDG receptor expression is developmentally regulated and the expression of the 8 members of this family of proteins appears to be tissue specific. EDG receptors couple to G-proteins to signal through Ras and MAPK pathways and other protein tyrosine kinase pathways. EDG-7 is also known as LPA3. The ligand for EDG-7 is lysophosphatidic acid (LPA), containing an sn-2 unsaturated fatty acid ester. Ligand binding activates Gq. EDG-7 is expressed by prostate and other tissues.
Lysophosphatidic acid receptor 3 (LPA3, EDG-7) is one of three lysophosphatidic acid (LPA) receptors (LPA1, LPA2, LPA3) that are members of large family of G-protein coupled receptors (GPCRs) that also include those for sphingosine-1-phosphate (S1P1-5) (1). The LPA receptors mediate many cellular responses including cytoskeletal rearrangements, cell proliferation, and inhibition of gap junction communication (2,3). Murine and human LPA3 have 353 amino acids with an estimated molecular weight of 40kD (4,5). The mRNA level of LPA3 is high in testes, kidney, and lung but low in intestine, heart, thymus, and stomach (5).
